preparation | under nitrogen protection, 2, 4-dimethoxypyrimidine (1.4g,10mmol) is dissolved in tetrahydrofuran (14mL), cooled to about 0 ℃, and 1M magnesium dichloride (2,2,6,6-tetramethylpiperidine) lithium salt (11mL,11mmol) is added dropwise under stirring, continue stirring for 2 hours, then add iodine (2.79g,11mmol) under stirring, maintain low temperature reaction for 2 hours, naturally return to room temperature, continue stirring for 6 hours. The reaction solution is poured into ammonium chloride aqueous solution for quenching, ethyl acetate is extracted three times, combined, washed, dried and concentrated. After the obtained crude column chromatography, 2, 4-dimethoxy-5-iodopyrimidine (2.34g, yield 88%) was obtained. |
